چکیده

context prenatal testing aims to identify fetal chromosomal and genetic disorders prior to delivery. current invasive procedures such as amniocentesis and chorionic villus sampling (cvs) pose a risk to mother and fetus and such diagnostic procedures are available only to high-risk pregnancies, which limits aneuploidy detection rate. the identification of cell-free fetal dna (cffdna) in maternal circulation has made noninvasive prenatal testing (nipt) possible. this review seeks to highlight the necessity of investing in nipt and briefly summarizes the technical aspects of the nipt and application of this method in clinical practice. evidence acquisition pubmed, ovid, scopus, and the cochrane database were searched for relevant articles published between 1995 and 2014, using appropriate keywords including prenatal screening, noninvasive testing, prenatal diagnosis, cell free fetal dna, maternal circulation, chromosomal aneuploidies, trisomy, and sex determination. results were restricted to systematic reviews, randomized clinical trials, meta-analysis, and observational studies. results the importance of prenatal diagnosis and risks associated with current invasive techniques makes nipt research morally and commercially beneficial. the outstanding advantages of nipt over current prenatal diagnosis techniques include increasing detection rate, enabling earlier diagnosis, and eliminating iatrogenic fetal loss and risk to the mother due to invasive procedures. at present, two major techniques for isolating cffdna, namely digital pcr and massively parallel sequencing (mps), have enabled the successful implementation of nipt into clinical practice such as fetal sex determination, rhd genotyping, and fetal chromosomal aneuploidy detection. conclusions the advent of new nipt using cffdna has been regarded as a revolution in prenatal testing and has attracted significant commercial interest in the field. it is not overoptimistic to predict that nipt will supplement or replace existing screening and diagnostic tools.

Cell-free fetal DNA in maternal plasma and noninvasive prenatal diagnosis.

The noninvasive nature of the detection of fetal DNA in the maternal circulation represents the greatest advantage over the conventional methods of prenatal diagnosis. The applications of this methodology involve the detection of the fetal sex, and diagnosis, intra-uterine treatment, and evaluation of the prognosis of many diseases. Fetal cells detected in the maternal circulation have also bee...

Noninvasive prenatal test for fetal chromosomal aneuploidies by massively parallel sequencing of cell- free fetal DNA in maternal plasma: The first clinical experience in Korea

mostly trisomy 21, relies exclusively on biochemical and sonographic measurements performed in the first and second trimesters. With a 3-5% false-positive rate, first-trimester screening achieves a detection rate of about 60-95% for trisomy 21 [1-3]. In 1997, Lo et al. [4] first demonstrated the presence Noninvasive prenatal test for fetal chromosomal aneuploidies by massively parallel sequenci...

Comparison of noninvasive prenatal testing of cell‐free DNA in maternal blood and amniocentesis for evaluation of aneuploidy

Background: The aim of this study was to compare noninvasive prenatal testing (NIPT) of cell‐free DNA in maternal blood and amniocentesis in the diagnosis of aneuploidy. This study was designed to evaluate sensitivity, specificity, accuracy, positive predictive value and negative predictive value of NIPT for detection of aneuploidies compared gold standard test of amniocentesis. Cell-free fetal DNA in the maternal serum and plasma: current and evolving applications.

PURPOSE OF REVIEW Free fetal nucleic acids, found in the plasma of every pregnant woman, have made a substantial impact on prenatal diagnosis. The past decade has seen the introduction of routine noninvasive prenatal diagnosis (NIPD) using DNA extracted from maternal plasma for a number of clinical complications of pregnancy, notably feto-maternal blood group incompatibility, fetal sexing and e...

Initial clinical laboratory experience in noninvasive prenatal testing for fetal aneuploidy from maternal plasma DNA samples

OBJECTIVE The aim of this study is to report the experience of noninvasive prenatal DNA testing using massively parallel sequencing in an accredited clinical laboratory. METHODS Laboratory information was examined for blood samples received for testing between February and November 2012 for chromosome 21 (Chr21), Chr18, and Chr13. Monosomy X (MX) testing was available from July 2012 for cysti...

Noninvasive Prenatal Diagnosis of Fetal Trisomy 18 and Trisomy 13 by Maternal Plasma DNA Sequencing

Massively parallel sequencing of DNA molecules in the plasma of pregnant women has been shown to allow accurate and noninvasive prenatal detection of fetal trisomy 21. However, whether the sequencing approach is as accurate for the noninvasive prenatal diagnosis of trisomy 13 and 18 is unclear due to the lack of data from a large sample set. We studied 392 pregnancies, among which 25 involved a...
